Base Pair Biotechnologies is pleased to provide this complete AptaColorTM test kit to easily and quickly demonstrate aptamer specificity in a common lateral flow assay format. Lateral flow assays are based on target immobilization and capillary transfer of detection reagents on a lateral flow strip. The AptaColorTM kit is a simple lateral flow assay designed to specifically detect a protein target with an aptamer developed by Base Pair Biotechnologies. The kit utilizes a biotin labeled aptamer and streptavidin gold conjugate solution that when combined with the target on the lateral flow strip yield a visible pink reaction. The assay demonstrates superb specificity of the aptamer by not binding a non target protein abundant in most biological assays. Directions for use: AptaColorTM Kit Materials Include:



Pulse centrifuge all tubes to accumulate liquids to bottom



Place strip into 50μ 50μL of the Biotinylated Aptamer solution in 2mL ( Tube A ) , within seconds the liquid will be ab-



Strip with immobilized Target and Non Target Proteins



( 5 0 μ L ) Biotinylated Aptamer solution ( Tube A )



( 5 0 μ L ) StreptavidinStreptavidin-gold conjugate solution ( Tube B )



2 X ( 50μ 50 μL ) PBS+ solution ( Tube C and D )

Once absorbed transfer strip to 2mL ( Tube B ) containing 50μ 50μL of the StreptavidinStreptavidin-gold conjugate solution which will also be absorbed into the strip



Move strip to 2mL ( Tube C ) and rinse for 10 sec.with 50μ 50μL PBS+ solution, repeat with another 50μ 50μL of PBS+ in 2mL ( Tube D )



Strip should turn pink where the Target Protein has been immobilized ( T ) and not turn pink where the Non Target Protein has been immobilized ( NT ) , dispose of all reagents in a biohazard container.
