PLANT BIOLOGY ANTIMICROBIALS Antimicrobials are often used in plant tissue culture applications to control microbial infection (bacterial, viral, and fungal), select for generically transformed plants, and to eliminate Agrobacterium after successful transformation. Choosing the correct antimicrobials to use in plant cell culture can be challenge due to their toxicity to various plant tissues. The antimicrobials listed below have low toxicity to plant tissue and are recommended for use in plant tissue culture applications. Bialaphos sodium

Timentin

Bialaphos sodium has successfully been used in major cereal species transformation including (wheat, rice, maize, barley, sorghum, oat and rye). The ‘bar’ gene was in these cases successfully incorporated in the plant genome, together with the gene of interest.

Timentin is used in Agrobacterium mediated transformations to control Agrobacterium growth. Carbenicillin and cefotaxime can be used in the same applications, however, a study by Nauerby et. al. (1997) found timentin to be less toxic.

Carbenicillin

Cefotaxime is often used in Agrobacterium tumefaciens mediated transformations to manipulate bacterial growth. It is also sometimes used in combination with Vancomycin as it has a synergistic effect.

Carbenicillin disodium is a beta-lactam antibiotic that is often used to select for cells that have been transformed with a plasmid containing the ampR gene which confers resistance to ampicillin and carbenicillin.

Hygromycin B Hygromycin B is an aminoglycoside antibiotic and is commonly used to select for transfected cells containing the hyg or hph resistance genes.

Streptomycin sulfate is an aminoglycoside antibiotic and inhibits protein synthesis in Gram-positive and Gram-negative bacteria.
