Upgrade to ÄKTA™ avant: 10 ways to boost productivity By upgrading from ÄKTAexplorer to ÄKTA avant chromatography system, you can improve productivity in process development. Depending on your application and how the system is operated, different ways to improve your productivity will have varying impact. Here we show 10 ways to boost your productivity.

1. Create methods fast and easy Quickly start your runs and spend less time on troubleshooting. Processes are easily created in the UNICORN™ system control software using predefined methods for a wide range of different chromatographic techniques. Methods can also be created by simply dragging and dropping phases reflecting individual steps of a chromatography run.

2. Quickly prepare your buffers in-line Your choice of buffer is conveniently mixed in-line using BufferPro. The tool minimizes your effort by accurately delivering buffers of desired composition for a large number of applications, such as automatic scouting for a large number of pH and conductivity conditions to find optimal buffer compositions.

3. Save time with integrated design of experiments (DoE) Using a DoE approach, the number of experiments required to obtain sufficient process information can be greatly reduced. The UNICORN system control software has an integrated DoE functionality. The software allows you to set up your experimental design and automatically generate chromatography scouting runs to save time when designing experiments and eliminates the need for data transfer between UNICORN and an external DoE software.

Automated methods can quickly and easily be created by dragging and dropping in Method Editor of the UNICORN software.

4. Seamless process scaling

Scaling of your protein purification process demands easy technology transfer between scales. In the UNICORN software, methods are quickly scaled using a time-saving functionality that automatically scales methods between ÄKTA avant 25 and ÄKTA avant 150. Suitable for use with ÄKTA avant 150, AxiChrom columns give you access to proven process scalability from ÄKTA avant up to the larger-scale ÄKTApilot and ÄKTAprocess chromatography systems.

Short runtimes are important, especially in manufacturing. ÄKTA avant can help reduce your runtimes by supporting high flows, and is well suited for use with high-flow chromatography media such as Capto™ ion exchange media.

ÄKTA avant supports higher flow rates than ÄKTAexplorer, enabling the use of high-flow chromatography media to shorten runtimes in your manufacturing facility. The bars represent the maximum obtained flow velocity for each system at the maximum flow rate for different column diameters. The right hand scale shows the flow velocity translated into residence time for a 20 cm bed height column, with typical residence times for three common process steps highlighted.

6. Securely maximize flow rates For process times to be as short as possible, the chromatography run is often performed at the maximum flow rate of the packed column. To enable unattended operation under such conditions, pressure sensors before and after the column help maintain column pressure below its limit to protect the column and packed bed from overpressure.

7. Take control over your columns With ÄKTA avant, the Column Logbook keeps track of every individual column used in your lab and provides column history and performance trending data. With the Column Logbook, you can maintain columns based on performance rather than according to a fixed schedule, eliminating unnecessary maintenance runs and might even prolong column lifetime.

8. Minimize hassle and hands-on time The high degree of automation reduces time for manual interaction with the system. System features, such as automatic switching between sample pump and loop; in-line pH electrode calibration, bypass, and storage; and automatic tube type detection by fraction collector, minimize hassle. For your convenience, air sensors allow sample to be completely loaded and the process to automatically continue to the next step without air being introduced into the flow path.

9. Optimize instrument use

In-line calibration, bypass, and storage of the pH electrode eliminates many of the hassles of traditional pH valves, with which the same operation is performed offline.

For increased instrument utilization, purification tasks can easily be distributed between available instruments with ÄKTA avant installed in a network configuration. Methods can be created and processes monitored from a remote office computer. The network capability enables data to be securely stored on a common data base server, and simplifies teamwork by facilitating method transfer and sharing.
